Comparison of quality differences of flavonoids in Sophora japonica L. flowers and Robinia pseudoacacia L. flowers

Abstract: Sophora japonica L. flowers and Robinia pseudoacacia L. flowers are closely related varieties, both of which have medicinal and edible value and are rich in flavonoids. The standard of Sophora japonica L. flowers in pharmacopoeia was used to explore the difference of flavonoids between the two, so as to facilitate the distribution and clinical distinction. With rutin as the control, TLC qualitative comparison, colorimetry determination of total flavonoids content and HPLC determination of rutin content were conducted for the two sophora flower. The difference between the chromatograms of Sophora japonica L. flower and Robinia pseudoacacia L. flower was very obvious. Rutin was the main chemical composition in Sophora japonica L. flower, and there were different principal components from rutin in Robinia pseudoacacia L. flower. The determination results showed that the contents of total flavonoids and rutin of Sophora japonica L. flowers were about 1.5 times and 5~10 times of that of Robinia pseudoacacia L. flowers, respectively. The total flavonoids content of the two kinds of Sophora japonica L. flowers was greater than 8%; the total flavonoids of the same species of Sophora japonica L. flowers from different origins showed differences; after processing, the contents of total flavonoids and rutin gradually decreased in the order of raw Sophora japonica L. flowers, fried Sophora japonica L. flowers and Sophora japonica L. flowers charcoal. The chromatographic differences between the two kinds of Flos Sophorae were obvious, which could be used for the qualitative identification of the two; the contents of total flavonoids and rutin in Robinia pseudoacacia L. flowers were significantly lower than those in Sophora japonica. L. flowers There were differences in the amount of the same species of Sophora japonica L.flowers from different habitats, and there was little difference in chromatogram. The flavonoids in Flos Sophorae after processing changed to some extent; the results can be used for the comparison of flavonoids differences between the two.
