Study on extraction process of polysaccharide in Codonopsis pilosula based on response surface methodology

Abstract: The polysaccharide in Codonopsis pilosula was extracted by water extraction and alcohol precipitation. The effects of reflux time, extraction temperature and liquid-solid ratio on the extraction rate of the polysaccharide in Codonopsis pilosula were investigated by the single factor test. The extraction process was optimized by Box-Behnken response surface methodology. The optimal extraction process was liquid-solid ratio of 11.2∶1(mL/g), extraction temperature of 80 ℃ and reflux time of 1.5 h. Under these conditions, the yield of Codonopsis pilosula polysaccharide was 19.38%, and the measured value was 0.573 percentage points lower than the theoretical value of the model, with a good fitting degree, which fully verified the accuracy of the model. The process optimization was reasonable and effective.
