黄精内生细菌的分离及鉴定

Separation and identification of endophytic bacteria from Polygonatum sibiricum

摘要: 以陇东子午岭自然保护区采挖的鸡头黄精(Polygonatum sibiricum)为材料,通过组织块分离法和琼脂打孔法从其块茎中分离到1株对金黄色葡萄球菌(Staphylococcus aureus)有抑菌作用且遗传稳定的内生细菌H-3,并通过基因组DNA提取、16S rDNA基因克隆测序分析及形态学特征,得出该菌为环状芽孢杆菌(Bacillus circulans)。对其发酵液醇提固形物多糖测定显示,其多糖含量达24.00%。

Abstract: Using Polygonatum sibiricum collected from Ziwuling Nature Reserve in Longdong as material, an endophytic bacterium H-3 with antibacterial activity and genetic stability against Staphylococcus aureus was isolated from its tuber by tissue block separation and agar drilling method. The strain was identified as Bacillus circulans by genomic DNA extraction, 16S rDNA gene cloning and sequencing analysis and morphological characteristics. The determination of solid polysaccharide extracted from fermentation broth showed that the polysaccharide content reached 24.00%.
