Extraction and determination of antioxidant activity in vitro of total flavonoids from Chenopodium formosanum Koidz seeds

Abstract: Taking the seeds of Chenopodium formosanum Koidz as raw materials, the orthogonal test method was used to explore the effects of enzyme mass fraction, solid-liquid ratio, enzymolysis temperature and ultrasonic time on the extraction of Chenopodium formosanum Koidz seeds, and the antioxidant activity in vitro was determined by analyzing the scavenging ability of flavonoids from Chenopodium formosanum Koidz seeds to DPPH· and ·OH. The results showed that the best technological condition for extracting flavonoids from the seeds of Chenopodium formosanum Koidz was cellulase with the mass fraction of 0.6%, the solid-liquid ratio of 1∶40, the enzymatic hydrolysis temperature of 60 ℃, and the ultrasonic time of 10 min. Under this condition, the extraction rate of flavonoids was as high as 27.81 mg/g. In vitro antioxidant test showed that the flavonoids extracted from the seeds of Chenopodium formosana Koidz had 92.91% and 94.72% scavenging activity on DPPH· and ·OH, respectively, and had certain antioxidant activity.
