Study on inducing callus of pear (Pyrus communis L. ‘Conference') by orthogonal array-based method

Abstract: Pyrus communis L. ‘Conference' was used as material to induce callus, and L9(34) orthogonal design was used to explore the effects of various factors on callus induction of ‘Conference' pear.The results showed that the order of influence of each factor on callus induction: TDZ concentration > explants type > IBA concentration, and the influence of TDZ and IBA on the experimental results was significant (P<0.05), in addition, there were significant differences among different levels. A1B3C3 proved to be the best combination, stem segments were used as explants, MS+2.0 mg/L TDZ +0.5 mg/L IBA medium and dark culture for 35 days were adopted, and the induction rate was 100%.
