Study on the interaction of porcine APOA2 promoter and transcription factor GATA1

Abstract: This study was aimed to clone the potential promoter region of porcine APOA2 gene and analyze its transcriptional activity,obtain its core promoter region,and analyze the effect of transcription factor GATA1 on the transcriptional activity of APOA2 gene for further studying the function of APOA2 gene in meat traits.The results showed that APOA2 gene was mainly expressed in liver and adipose tissue of white pigs.The results of dual luciferase activity indicated that there was the core promoter region of APOA2 gene5 'flanking region at -813 to -565 bp,and there were more likely to be some cis-acting elements promoting APOA2 transcription in the region of -659 to -565 bp.The results of GATA1 over-expression indicated that transcription factor GATA1 promoted the transcription of APOA2 gene.Site-directed mutagenesis assay confirmed that transcription factor GATA1 could directly bind to GATA1 binding sites (-608 to -599 bp) in the promoter region of APOA2 gene.Taken together,these results suggested that transcription factor GATA1 could directly target the promoter region of APOA2 gene and promoted APOA2 transcription.
