Response surface methodology to optimize the ultrasonic-assisted extraction process of Ganoderma lucidum flavonoids

Abstract: The experiment used response surface methodology to optimize the ultrasonic-assisted extraction of flavonoids from Ganoderma lucidum. The effects of four factors, including ethanol concentration, extraction time, solid-liquid ratio, and ultrasonic power, on the extraction yield of flavonoids in Ganoderma lucidum were investigated. Using flavonoids rate as the response value, the Box-Behnken center design developed mathematical model for response surface analysis. The results showed that the optimal extraction process parameters of Ganoderma lucidum flavonoids were ethanol concentration of 86%, solid-liquid ratio of 1:20 (g/mL), extraction power of 185 W, and extraction time of 30 min. The mathematical model established under these conditions was verified by the test. The actual extraction yield of Ganoderma lucidum flavonoids was 7.55%. The error with the predicted value of 7.62% was less than 3%, thus confirming that the flavonoids extraction process parameters obtained with Design-Expert 8.0.6 software were real and reliable, and had practical application value.
