Study on optimization of extraction process of Bupleurum chinensis saponins by central composite design-response surface methodology

Abstract: On the basis of single factor experiment, the effect of extraction times on the content of saponins in Bupleurum chinense was analyzed. The extraction process of saikosaponins was optimized by response surface method of central composite design. The effects of extraction time, extraction temperature and the ratio of liquid to material on saikosaponins were investigated. The results showed that the best conditions of total saponins of Bupleurum chinense were extraction temperature 80 ℃, extraction time 60 min, liquid-to-material ratio 8∶1(mL∶g), and extraction twice. Under the optimal condition, the content of total saponins of Bupleurum chinense was 1.80 mg/g, which was well fitting with the predicted value of 1.81 mg/g. The extraction process by response surface methodology is accurate, reliable and predictable.
