广西玉林肉桂炭疽病病原菌鉴定及生物学特性

Identification and biological characteristics of the pathogen causing anthracnose on Cinnamomum cassia in Yulin, Guangxi

摘要: 为明确广西玉林市肉桂(Cinnamomum cassia)炭疽病的病原菌种类及其生物学特性。采用组织分离法从病叶中分离纯化病原菌,通过致病性测定、形态学观察及基于4个基因片段(ITSACTGAPDHTUB2)的联合系统发育分析对病原菌进行鉴定,并研究碳源、氮源、温度、光照和pH对病原菌生长的影响。结果表明,引起玉林市肉桂炭疽病的病原菌为果生刺盘孢(Colletotrichum fructicola),本研究首次报道该病原菌侵染肉桂。该菌株的最适生长碳源为麦芽糖,最适氮源为磷酸二氢铵和蛋白胨,最适生长温度为28 ℃,在连续光照及pH 6的条件下生长最佳,致死条件为52 ℃处理15 min。

Abstract: To clarify the pathogen species causing anthracnose on Cinnamomum cassia in Yulin, Guangxi, and its biological characteristics, the pathogen was isolated and purified from diseased leaves using the tissue isolation method. It was identified through pathogenicity testing, morphological observation, and concatenated phylogenetic analysis based on four gene fragments (ITS, ACT, GAPDH, and TUB2). The effects of carbon sources, nitrogen sources, temperature, light, and pH on mycelial growth were also investigated. The results showed that the pathogen causing anthracnose on Cinnamomum cassia in Yulin was Colletotrichum fructicola. This study reported for the first time that Colletotrichum fructicola infected Cinnamomum cassia. The optimal carbon source for mycelial growth was maltose, and the optimal nitrogen sources were ammonium dihydrogen phosphate and peptone. The optimal temperature for growth was 28 ℃, with the best growth observed under continuous light and at pH 6. The lethal condition for the mycelia was treatment at 52 ℃ for 15 min.
